Shan Masood remained unbeaten on 88 off 137 balls, hitting 11 fours, in a 155-run partnership with Imam-ul-Haq, who scored 63.
The Pakistan innings had a difficult start, with opener Azan Awais bowled by Kemar Roach for a nine-ball duck.
Salman Ali Agha was unbeaten on nought off 9 balls at stumps.
West Indies were dismissed for 311 in their first innings, with Mohammad Ali taking four wickets for Pakistan.
Pakistan will resume their innings on day three, still 112 runs behind.
